IMMUNOLOGY AND MOLECULAR BIOLOGY—PS 4165 A Plasmid DNA Encoding Chicken Interleukin-6 and Escherichia coli K88 Fimbrial Protein FaeG Stimulates the Production of Anti-K88 Fimbrial Antibodies in Chickens

چکیده
Immunization using a plasmid to deliver an encoded protein for expression in situ as the antigen is a promising technology. A plasmid encoding the enterotoxigenic Escherichia coli K88 fimbrial protein FaeG when injected into chickens stimulates the production of antibodies against the fimbrial protein, similar to what has been observed in mice. The efficacy of a genetic adjuvant on fimbrial antibody production was tested by introducing the gene for chicken interleukin-6 in tandem with the faeG gene. Expression of both the fimbrial FaeG protein and chicken interleukin-6 protein was confirmed in COSM6 cells. Slightly higher antiFaeG antibody titer in chick(

Construction of a Novel DNA Vaccine Candidate encoding LmSTI1-PpSP42 Fusion Protein from Leishmania major and Phlebotomus papatasi against Cutaneous Leishmaniasis
Background: Cutaneous leishmaniasis (CL) is a serious public health problem in many tropical countries. The infection is caused by a protozoan parasite of Leishmania genus transmitted by Phlebotominae sandflies. In the present study, we constructed a eukaryotic expression vector to produce a fusion protein containing LmSTI1 from Leishmania major (L. major) and PpSP42 from Phlebotomus papatasi (...
متن کاملThe Increase in Protein and Plasmid Yields of E. coli with Optimized Concentration of Ampicillin as Selection Marker
Background: Escherichia coli is still the common host for ing and heterologous protein expression. Various strategies have been employed to increase protein expression in E. coli, but, it seems that external factors such as selection marker concentration can drastically aff ect the yield of protein and plasmid. Objectives: Alterations of protein expression...
متن کاملCloning and Expression of the Variable Regions of Anti-EGFR Monoclonal Antibody in E. coli for Production of a Single Chain Antibody
Background:Epidermal growth factor receptor (EGFR) overexpression is a characteristic of several malignancies and could be considered as an excellent target for designing specific inhibitors such as anti-EGFR monoclonal antibodies for cancer therapy. Drawbacks exerted by large sizes of full-length antibodies have lead to the development of single chain antibodies, which benefit from having smal...
متن کاملOptimization of Cloning Conditions for high-level Production of Recombinant Mouse Interleukin-2 in Escherichia coli
Backgrounds and objectives: Interleukin 2 (IL-2) secreted by activated CD4+ T cells has been known as a major mediator in both adaptive and native immune system due to a board range of effects on different cells in the immunity system (1-6). Methods: cDNA synthesis was performed using gene- specific primers designed by Gene Runner software after RNA extraction of mouse splenocytes. PCR pro...

Background & Objectives: Antisense peptide nucleic acids (PNA) that target growth essential genes show potent bactericidal properties without cell lysis. We considered the possibility that whether PNA treatment influence the bacteria total nucleic acids content and apply approach to develop a new delivery system to Dendritic cells (DCs). DCs are the most potent antigen presenting cells in th...
